Introduction
Have you ever wondered how scientists extract DNA from an organism? All living organisms have DNA, which is short for deoxyribonucleic acid; it is basically the blueprint for everything that happens inside an organism?s cells. Overall, DNA tells an organism how to develop and function, and is so important that this complex compound is found in virtually every one of its cells. In this activity you?ll make your own DNA extraction kit from household chemicals and use it to separate DNA from strawberries.?

Background
Whether you?re a human, rat, tomato or bacterium, each of your cells will have DNA inside of it (with some rare exceptions, such as mature red blood cells in humans). Each cell has an entire copy of the same set of instructions, and this set is called the genome. Scientists study DNA for many reasons: They can figure out how the instructions stored in DNA help your body to function properly. They can use DNA to make new medicines or genetically modify crops to be resistant to insects. They can solve who is a suspect of a crime, and can even use ancient DNA to reconstruct evolutionary histories!

To get the DNA from a cell, scientists typically rely on one of many DNA extraction kits available from biotechnology companies. During a DNA extraction, a detergent will cause the cell to pop open, or lyse, so that the DNA is released into solution. Then alcohol added to the solution causes the DNA to precipitate out. In this activity, strawberries will be used because each strawberry cell has eight copies of the genome, giving them a lot of DNA per cell. (Most organisms only have one genome copy per cell.)

Materials

  • Rubbing alcohol
  • Measuring cup
  • Measuring spoons
  • Salt
  • Water
  • Dishwashing liquid (for hand-washing dishes)
  • Glass or small bowl
  • Cheesecloth
  • Funnel
  • Tall drinking glass
  • Three strawberries
  • Resealable plastic sandwich bag
  • Small glass jar (such as a spice or baby food jar)
  • Bamboo skewer, available at most grocery stores. (If you use a baby food or short spice jar, you could substitute a toothpick for the skewer.)

Preparation

  • Chill the rubbing alcohol in the freezer. (You?ll need it later.)
  • Mix one half teaspoon of salt, one third cup of water and one tablespoon of dishwashing liquid in a glass or small bowl. Set the mixture aside. This is your extraction liquid. Why do you think there is detergent in the extraction liquid?
  • Completely line the funnel with cheesecloth. Insert the funnel tube into the tall drinking glass (not the glass with the extraction liquid in it).
  • Remove and discard the green tops from the strawberries.

Procedure

  • Put the strawberries into a resealable plastic sandwich bag and push out all of the extra air. Seal the bag tightly.
  • With your fingers, squeeze and smash the strawberries for two minutes. How do the smashed strawberries look?
  • Add three tablespoons of the extraction liquid you prepared to the strawberries in the bag. Push out all of the extra air and reseal the bag. How do you think the detergent and salt will affect the strawberry cells?
  • Squeeze the strawberry mixture with your fingers for one minute. How do the smashed strawberries look now?
  • Pour the strawberry mixture from the bag into the funnel. Let it drip through the cheesecloth and into the tall glass until there is very little liquid left in the funnel (only wet pulp remains). How does the filtered strawberry liquid look?
  • Pour the filtered strawberry liquid from the tall glass into the small glass jar so that the jar is one quarter full.
  • Measure out one half cup of cold rubbing alcohol.
  • Tilt the jar and very slowly pour the alcohol down its side. Pour until the alcohol has formed approximately a one-inch-deep layer on top of the strawberry liquid. You may not need all of the one half cup of alcohol to form the one-inch layer. Do not let the strawberry liquid and alcohol mix.
  • Study the mixture inside of the jar. The strawberry DNA will appear as gooey clear/white stringy stuff. Do you see anything in the jar that might be strawberry DNA? If so, where in the jar is it?
  • Dip the bamboo skewer into the jar where the strawberry liquid and alcohol layers meet and then pull up the skewer. Did you see anything stick to the skewer that might be DNA? Can you spool any DNA onto the skewer????
  • Extra: You can try using this DNA extraction activity on lots of other things. Grab some oatmeal or kiwis from the kitchen and try it again! Which foods give you the most DNA?
  • Extra: If you have access to a milligram scale (called a balance), you can measure how much DNA you get (called a yield). Just weigh your clean bamboo skewer and then weigh the skewer again after you have used it to fish out as much DNA as you could from your strawberry DNA extraction. Subtract the initial weight of the skewer from its weight with the DNA to get your final yield of DNA. What was the weight of your DNA yield?
  • Extra: Try to tweak different variables in this activity to see how you could change your strawberry DNA yield. For example, you could try starting with different amounts of strawberries, using different detergents or different DNA sources (such as oatmeal or kiwis). Which conditions give you the best DNA yield?

There was once an old adage in the auto industry, ?safety doesn?t sell.? Don?t try to convince today?s motorists.? Safety has, in fact, become one of the highest-ranked attributes for most car shoppers, whether they?re looking for a minivan or sports car.

So, a new study by Insure.com is likely to deliver more than a few surprises when it comes to listing the safest and least safe among 750 different vehicles now on the market.? Some of the results might seem intuitive, especially to those who equate size with safety.? The pint-sized Fiat 500 is the least-safe vehicle on the list, according to the study which was produced by Insure.com in cooperation with the Insurance Institute for Highway Safety.? On the other hand, the GMC Sierra 1500 pickup was the top-ranked model.?

"If safety is a priority, you should avoid the smallest cars," said IIHS spokesman Russ Rader. "Weight counts. Smaller, lighter cars are safer than they used to be, but all things being equal, people riding in bigger, heavier vehicles get more protection in crashes."

But there are also some surprises.? While big GMC models took four of the five ?safest? slots, the four-door Porsche Cayenne ranked second.? On the other hand, the Mercedes-Benz passenger car flagship, the premium CL 500 coupe was fifth on the list of least-safe vehicles.?

From a brand standpoint, General Motors scored particularly well ? as noted, the GMC brand landing four of the top five slots.? In other words, being a best-seller in your segment doesn?t always guarantee buyers will get the best in terms of safety.

The study might generate some controversy as there a variety of ways to measure safety.? Take the Fiat 500 which has actually generated very good scores in IIHS crash tests with its integrated safety cage and airbags.

Back when the PlayStation 2 was the world's most popular game console, the original Xbox was struggling to gain a post-Halo audience and the GameCube was a cute diversion with a couple decent titles, I trolled gaming message boards. I was so sure that anyone who bought anything other than a PS2 was out of their mind that I set on the most immature mission in history: to inform the masses, proselytize the word as handed down by Hirai-san, and ridicule those who were wasting their time with The Wind Waker.

Those who were active gamers at the time probably read that paragraph and are now either annoyed or cracking a wry smile in understanding. The latter are the ones who played Ico. Those who weren't gamers at the time can insert their own products into the mix for context.

Congress passes $50.5B Superstorm Sandy aid bill

WASHINGTON (AP) ? Three months after Superstorm Sandy ravaged coastal areas in much of the Northeast, Congress on Monday sent a $50.5 billion emergency relief measure for storm victims to President Barack Obama for his signature.

"I commend Congress for giving families and businesses the help they deserve, and I will sign this bill into law as soon as it hits my desk," Obama said in a statement late Monday.

Despite opposition from conservatives concerned about adding billions of dollars more to the nation's debt, the Senate cleared the long-delayed bill, 62-36, after House Republicans had stripped it earlier this month of spending unrelated to disasters. All 36 votes against the bill were from Republican senators.

"This is a huge relief," said Sen. Charles Schumer, D-N.Y., noting the vote came 91 days after Sandy struck.

The House passed the bill two weeks ago. Lawmakers say the money is urgently needed to start rebuilding homes, businesses, public transportation facilities and other infrastructure damaged by the Oct. 29 storm, one of the worst to strike the Northeast.

Sandy roared up the East Coast and is blamed for more than 130 deaths and tens of billions of dollars in property damage, particularly in New York and New Jersey. The measure is aimed primarily at helping residents and businesses as well as state and local governments rebuild.

The biggest chunk of money is $16 billion for Housing and Urban Development Department community development block grants. Of that, about $12.1 billion will be shared among Sandy victims as well as those from other federally declared disasters in 2011-2013. The remaining $3.9 billion is solely for Sandy-related projects.

More than $11 billion will go to the Federal Emergency Management Agency's disaster relief aid fund for providing shelter, restoring power and other storm-interrupted utility services and meeting other immediate needs arising from Sandy and other disasters. Another $10 billion is devoted to repairing New York and New Jersey transit systems and making them more resistant to future storms.

Sen. Dan Coats, R-Ind., said Republicans weren't seeking "to undermine" help for Sandy victims but instead were trying to make sure that the money was actually being spent on emergency needs.

"We're simply trying to say we need some standards," Coats said.

Earlier in January, Congress approved and Obama signed a $9.7 billion bill to replenish the National Flood Insurance Program, which has received well over 100,000 flood insurance claims related to Sandy. Added to the new, $50.5 billion package, the total is roughly in line with the $60.4 billion that Obama requested in December.

The aid package was greased for passage before the last Congress adjourned and the new one came in on Jan. 3. But Speaker John Boehner refused to bring it to the floor after two-thirds of House Republicans voted against a "fiscal cliff" deficit-reduction deal raising taxes on couples making more than $450,000 a year while deferring some $24 billion in spending cuts to defense and domestic programs.

The ruckus after the Senate had passed an earlier $60.4 billion Sandy relief package by a nearly 2-to-1 margin on Dec. 28 exposed deep political divisions within Republican ranks. "There's only one group to blame for the continued suffering of these innocent victims, the House majority and their speaker, John Boehner," Republican New Jersey Gov. Chris Christie fumed at the time.

Top House Republicans responded by bringing new Sandy aid legislation to the floor under ground rules designed to win over as many Republicans as possible while retaining support from Democrats eager to approve as much in disaster aid as possible.

GOP leaders cut spending in the Senate bill unrelated to disasters. One would have transferred $1 billion for training Iraqi policemen to instead be used to bolster security at U.S. diplomatic missions. The shift in money followed a Sept. 11 terrorist attack on the American consulate in Benghazi, Libya, where the U.S. ambassador and three other Americans were killed.

Also deleted were $188 million for an Amtrak expansion project that included new, long-planned tunnels from New Jersey to Penn Station in Manhattan, and another $150 million for fisheries disasters that states such as Alaska and Mississippi could have shared.

After all the cost-cutting, 179 House Republicans still voted against the disaster aid package with only 49 favoring it. GOP leaders had to rely on yes votes from 192 Democrats to pass it.

As with past natural disasters, the Sandy aid bill is not offset with spending cuts, meaning the aid adds to the deficit. The lone exception is an offset provision requiring that $3.4 billion for Army Corps of Engineers projects to protect against future storms be covered by an equal amount of unspecified spending cuts in other programs before next October.

The Senate on Monday rejected, 35-62, an attempt by Sen. Mike Lee, R-Utah, to amend the final package Monday with a provision to cut federal programs across the board by one-half of 1 percent through 2021 as a way to prevent the disaster aid from swelling the U.S. debt.

As of Monday, FEMA said it spent $3.3 billion in disaster relief money for shelter, restoring power and other immediate needs arising from the storm.

New York, New Jersey, Connecticut, West Virginia, Virginia, Maryland, New Hampshire, Ohio, Delaware, Rhode Island, Pennsylvania, Massachusetts and the District of Columbia have shared that money.

Mission Creep in Mali? Britain's Growing Military Commitment

Britain?s growing military commitment to Mali and West Africa is likely to pass 400 within a matter of weeks, sparking fears of ?mission creep? and cross-party criticism that UK forces are being sucked into a Vietnam-style conflict without a defined exit strategy.

A fortnight ago David Cameron promised that Britain would only send ?tens, not hundreds? of troops to help the French-led operation. That prediction from the Prime Minister has been shredded as UK assistance to the French-led operation has significantly increased.

The growing importance of the unrest in West Africa was highlighted last night when Downing Street announced the PM would be flying to Algeria tomorrow for urgent talks with Prime Minister, Abdelmalek Sellal.

Mr Cameron was already scheduled to attend a development conference in Monrovia, Liberia, but added the Algerian meeting following the recent hostage crisis at the Amenas gas plant which left six Britons and 47 others dead.

Number 10 said the discussions were likely to include the military campaign in Mali, a neighbour-state to Algeria.

The Defence Secretary Phil Hammond was forced to make a statement to the Commons today admitting that troop numbers in the region would substantially rise with new support for an EU training mission in Mali and up to 200 additional personnel who will assist a United Nations-backed training programme for forces from neighbouring West African states.

UK military advisers are already in place helping the UN?s Africa-led International Support Mission to Mali (AFISMA). Ground and technical support teams for a Sentinel surveillance aircraft plus two C-17 transport planes protected by combat-ready troops from the RAF Regiment, have also been deployed.

Around 20 British personnel are in the Malian capital, Bamako, liaising with French forces that have already passed the 3500 mark. Further British military assistance in the form of a specialist ferry to help land French armoured vehicles at ports in Senegal or The Gambia, alongside the offer of expert personnel to help staff a joint UK-French logistics headquarters in Mali , is also being considered.

Britain?s national security adviser, Sir Kim Darroch, has been in Paris this week holding talks with his French counterparts. The talks ? focused on France?s short-term military shopping list for Mali ? suggest the UK?s role could grow...

Well, Britannia's not ruling the waves these days, that's for sure. You'd think the country which once boasted the world's most powerful empire could handled deploying a few hundred commandos to Central Africa. Sheesh.

Almost 7 million birds are killed each year when they fly into communication towers that broadcast TV and radio and make cellphone conversations possible. Worse, the towers often kill birds that are already rare. So says a study in the journal Biological Conservation. [Travis Longcore et al, Avian mortality at communication towers in the United States and Canada: which species, how many, and where?]

For example, tower impacts kill more than 2,000 yellow rails per year. That's roughly 9 percent of the total population. Ninety-seven percent of all birds killed are songbirds, especially warblers. The red-eyed vireo suffers some of the biggest losses, some 581,000 deaths annually, though that represents less than 1 percent of its population.

The Southeast and Midwest lead the country in tower-bird collisions. That's because these regions have the largest concentrations of the tallest towers, up to 900 feet high. While all of the more than 80,000 communication towers in North America cause problems, the roughly 1,000 tallest towers cause 70 percent of the bird deaths, luring birds to their doom with red warning lights that are always on.

A partial solution is relatively simple: replacing the always-on red lights with blinking ones could cut the deaths by as much as 70 percent. Otherwise, Twitter could have a monopoly on tweets.

? ? ?Sometime later Paul said to Barnabas, ?Let us go back and visit the believers in all the towns where we preached the word of the Lord and see how they are doing.? Barnabas wanted to take John, also called Mark, with them, but Paul did not think it wise to take him, because he had deserted them in Pamphylia and had not continued with them in the work. They had such a sharp disagreement that they parted company. Barnabas took Mark and sailed for Cyprus, but Paul chose Silas and left, commended by the believers to the grace of the Lord. Acts 15:36-40

Dear Lord Jesus, I start this day grateful for the stories of brokenness and messiness included in the Scriptures. Thankfully, they weren?t edited out, for these kinds of stories give me hope. The Bible is quite authentic and unabashedly honest, about the mess we are in and the mess that we are. Mere men wouldn?t have included these kinds of broken relationships between your people, without your hand and heart guiding them.

In particular, I?m thankful today for this story of two good friends, Paul and Barnabas, having ?such a sharp disagreement that they parted company.? Lord, I?ve been in that situation, and right now I?m watching that story play out between two friends I deeply love and respect. It?s sticky and messy, and I hate being caught in the middle.

So I look to you right now for the resources of the gospel. I need wisdom from above; power in my weakness; and grace to love two stubborn, hurting people. My default mode is to not get involved?hoping the whole thing will just blow over or go away. But that?s not going to happen.

Jesus, the good news is you?ve come to destroy hostilities; to tear down walls of division; to make peace and to be our peace. You reconcile enemies to God and to one another, so I can trust you to be at work in this tense and complicated situation.

It took time plus grace for Paul and Barnabas to get back together. I don?t know what time plus grace will look like for my friends, but show me my role in the story you are writing. Help me listen from the heart and confront in love. Lead my praying and let me feel their hurt. Help me engage with both sides without taking sides. Help me stay present in the chaos without giving in to the pulls. As I serve my friends, show me more of my sin and more of the power of the gospel.

Jesus, no one is better at sticky-situation loving than you, so I abandon myself to you with my fears, my confusion, and my very real hope of your showing up. So very Amen I pray, with confidence, in your powerful and reconciling name.

Two years after David O. Russell?s The Fighter, the infamously controversial filmmaker is back with his latest blend of crappy people and hearts of gold.

Silver Linings Playbook is a romantic comedy, which is perfect for the holiday season and families looking for entertainment. But it also has characters with mental issues, which is perfect for Oscar season. Taking a break from Mark Wahlberg, Russell cast endearing, popular actors Bradley Cooper and Jennifer Lawrence as his protagonists.

After a stay in a mental institution for anger issues and being diagnosed as bipolar, Patrick (Cooper) moves back in with his parents (Robert De Niro and Jacki Weaver) when he discovers his wife has left him. Things at home don?t go too well, either: Pat?s dad also has a bad temper and OCD, causing conflict. Soon an old friend of Pat?s sets him up on a date with his wife?s sister Tiffany (Lawrence), who also has a bad attitude and suffered depression after her husband?s death. It?s a match made in therapy; Pat is in denial about the end of his marriage and asks Tiffany to help him win his wife back.

Chris Tucker and Julia Stiles co-star as Pat?s rehab pal and Tiffany?s sister.

Russell has experimented with his share of genres the last two decades, from the romantic comedy Flirting with Disaster (1996) to the war satire Three Kings (1999) to the quirky ensemble piece I Heart Huckabees (2004) to the boxing drama The Fighter (2010). Now he steps back to his romcom roots with his adaptation of Matthew Quick?s novel Silver Linings Playbook.

Playbook manages to be light and heartfelt while dealing with serious subjects like mental illness. Cooper, known primarily for comedies like Wedding Crashers (2004) and The Hangover (2009), disappears into Pat and delivers as the lead of a talented cast. Many already think Lawrence should receive an Oscar nomination for her portrayal of aggressive, loudmouthed Tiffany, and it?s easy to see why. She is as amusing and cute as she is frustrated and bitchy.

Silver Linings Playbook is charmingly offbeat in a way few filmmakers could succeed at.

WASHINGTON -- Former Republican vice presidential candidate Rep. Paul Ryan (Wis.) said Sunday that if Secretary of State Hillary Clinton were president, the nation's fiscal crisis would have been averted.

?Look, if we had a [Hillary] Clinton presidency, if we had Erskine Bowles as chief of staff of the White House or president of the United States, I think we would have fixed this fiscal mess by now," Ryan said on NBC's "Meet the Press." "[But] that?s not the kind of presidency we?re dealing with right now.? Bowles was chief of staff to former President Bill Clinton, and currently co-chairs the Campaign to Fix The Debt, a bipartisan deficit reduction effort.

The praise was especially noteworthy because speculation abounds in Washington over both Ryan and Clinton's presidential prospects in 2016, although neither has responded to questions about their future plans.

"We have to expand our appeal to more people and show how we'll take the country's founding principles and apply them to the problem of the day, solutions to fix our problems.

"We have to show our ideas are better at fighting poverty, better at solving health care, how our ideas are better at solving problems that people experience in their daily lives," he continued. "And that's a challenge we have to rise to, and I think we're up for it."

To that end, Ryan was quick to reject the notion that Republicans in the House were willing to shut down the government, which some of the most conservative members of the chamber had suggested could be a good thing at the height of the budget debates in December.

"We're not interested in shutting the government down," he said. "What happens on March 1 is spending goes down automatically. March 27 is ? the moment you're talking about, the continuing resolution expires. We are more than happy to keep spending at those levels going on into the future while we debate how to balance the budget, grow the economy, create economic opportunity. That's the kind of debate the country deserves."

Egypt deploys troops in Suez after 9 killed on anniversary of uprising

CAIRO/ISMAILIA, Egypt (Reuters) - Egypt's armed forces deployed troops in the city of Suez early on Saturday after nine people were shot dead during nationwide protests against President Mohamed Mursi, underlining the country's deep divisions as it marked the second anniversary of the uprising that toppled Hosni Mubarak.

Eight of the dead, including a policeman, were shot dead in Suez, and another was shot and killed in the city of Ismailia, medics said. Another 456 people were injured across Egypt, officials said, in unrest on Friday fuelled by anger at Mursi and his Islamist allies over what the protesters see as their betrayal of the revolution.

Mursi said the state would not hesitate in "pursuing the criminals and delivering them to justice". In a statement, he also called on Egyptians to respect the principles of the revolution by expressing their views peacefully.

The troops were deployed in Suez after the head of the state security police in the city asked for reinforcements. The army distributed pamphlets to residents assuring them the deployment was temporary and meant to secure the city.

"We have asked the armed forces to send reinforcements on the ground until we pass this difficult period," Adel Refaat, head of state security in Suez, told state television.

Friday's anniversary laid bare the divide between the Islamists and their secular rivals.

The schism is hindering the efforts of Mursi, elected in June, to revive an economy in crisis and reverse a plunge in Egypt's currency by enticing back investors and tourists.

Inspired by the popular uprising in Tunisia, Egypt's revolution spurred further revolts across the Arab world. But the sense of common purpose that united Egyptians two years ago has given way to internal strife that already triggered bloody street battles last month.

Thousands of opponents of Mursi massed on Friday in Cairo's Tahrir Square - the cradle of the revolt against Mubarak - to rekindle the demands of a revolution they say has been hijacked by the Muslim Brotherhood, the group from which Mursi emerged.

In Suez, the military deployed armored vehicles to guard state buildings, witnesses and security sources said, as symbols of government were targeted across the country.

Street battles erupted in cities including Cairo, Alexandria, Suez and Port Said. Arsonists attacked at least two state-owned buildings. An office used by the Muslim Brotherhood's political party was also torched.

"Our revolution is continuing. We reject the domination of any party over this state. We say no to the Brotherhood state," Hamdeen Sabahy, a popular leftist leader, told Reuters.

The Brotherhood decided against mobilizing for the anniversary, wary of the scope for more conflict after December's violence, stoked by Mursi's decision to fast-track an Islamist-tinged constitution rejected by his opponents.

The Brotherhood denies accusations that it is seeking to dominate Egypt, labeling them a smear campaign by its rivals.

'LEAVE! LEAVE! LEAVE!'

There were conflicting accounts of the lethal shooting in Suez. Some witnesses said security forces had opened fire in response to gunfire from masked men.

News of the deaths capped a day of violence that started in the early hours of Friday. Before dawn in Cairo, police battled protesters who threw petrol bombs and firecrackers as they approached a wall blocking access to government buildings near Tahrir Square.

Clouds of teargas filled the air. At one point, riot police used one of the incendiaries thrown at them to set ablaze at least two tents erected by youths, a Reuters witness said.

Skirmishes between stone-throwing youths and the police continued in streets around the square into the day. Ambulances ferried away a steady stream of casualties.

Protesters echoed the chants of 2011's historic 18-day uprising. "The people want to bring down the regime," they chanted. "Leave! Leave! Leave!" chanted others as they marched towards the square.

"We are not here to celebrate but to force those in power to submit to the will of the people. Egypt now must never be like Egypt during Mubarak's rule," said Mohamed Fahmy, an activist.

There were similar scenes in Suez and Alexandria, where protesters and riot police clashed near local government offices. Black smoke billowed from tires set ablaze by youths.

In Cairo, police fired teargas to disperse a few dozen protesters trying to remove barbed-wire barriers protecting the presidential palace, witnesses said. A few masked men got as far as the gates before they were beaten back.

Teargas was also fired at protesters who tried to remove metal barriers outside the state television building.

Outside Cairo, protesters broke into the offices of provincial governors in Ismailia and Kafr el-Sheikh in the Nile Delta. A local government building was torched in the Nile Delta city of al-Mahalla al-Kubra.

With an eye on parliamentary elections likely to begin in April, the Brotherhood marked the anniversary with a charity drive across the nation. It plans to deliver medical aid to one million people and distribute affordable basic foodstuffs.

Writing in Al-Ahram, Egypt's flagship state-run daily, Brotherhood leader Mohamed Badie said the country was in need of "practical, serious competition" to reform the corrupt state left by the Mubarak era.

"The differences of opinion and vision that Egypt is passing through is a characteristic at the core of transitions from dictatorship to democracy, and clearly expresses the variety of Egyptian culture," he wrote.

Mursi's opponents say he and his group are seeking to dominate the post-Mubarak order. They accuse him of showing some of the autocratic impulses of the deposed leader by, for example, driving through the new constitution last month.

"I am taking part in today's marches to reject the warped constitution, the 'Brotherhoodisation' of the state, the attack on the rule of law, and the disregard of the president and his government for the demands for social justice," Amr Hamzawy, a prominent liberal politician, wrote on his Twitter feed.

The Brotherhood says its rivals are failing to respect the rules of the new democracy that put the Islamists in the driving seat via free elections.

Woman sues Match.com for $10 million after brutal attack

A Las Vegas woman who was severely beaten by a man she met through Match.com is suing the online dating site for $10 million?two years and multiple surgeries after the attack that left her hospitalized for months.

The woman, Mary Kay Beckman, was stabbed multiple times with a butcher knife on Jan. 21, 2011?by?Wade Mitchell Ridley, and when?the knife broke, he stomped on her head. Ridley, who was sentenced to?28 to 70 years,?died in prison last year. He was also facing a murder charge in Arizona for the stabbing death of a former girlfriend a few weeks after the attack on Beckman.

Beckman filed suit in Clark County, Nevada,?accusing Match.com of negligence, negligent misrepresentation, deceptive trade, failure to warn and negligent infliction of emotional distress.

The site, she said, failed to warn her about the dangers of meeting "an individual whose intentions are not to find a mate, but to find victims to kill or rape."

The real estate agent said she joined Match.com about a month before her first in-person meeting with Ridley on Sept. 26, 2010. They dated for 10 days, but she called it off. That's when Ridley started sending her threatening and harassing messages.?

On Jan. 21, 2011, Ridley attacked Beckman in her garage, and left her for dead, she says, when she stopped making a "gurgling noise."?

Beckman, now 50, continues to recover and to speak out against online dating?"I do not believe that online dating is a safe venue for men or women,"?she?recently?told?a?local?FOX?TV?reporter.

Match.com, in a statement to NBC News Friday, said that what happened to Beckman "is horrible, but this lawsuit is absurd.?The many millions of people who have found love on Match.com and other online dating sites know how fulfilling it is. And while that doesn't make what happened in this case any less awful, this is about a sick, twisted individual with no prior criminal record, not an entire community of men and women looking to meet each other."

Safety, the site said, "is very important to Match," which, like many other online dating sites, includes online and offline tips for staying safe.

In California, Match.com and two other dating sites, eHarmony and Spark Networks, signed a joint statement of business principles, agreeing to screen for sex offenders and take other safety steps after a woman was assaulted on a date, the state attorney general's office said last March.?

The joint statement ?was prompted by the 2010 sexual assault of a Los Angeles-area woman by a man she met through Match.com, a spokeswoman for the state attorney general's office said. The woman sued Match.com, seeking a court order requiring the site to check applicants' backgrounds to weed out convicted sex offenders. She dropped the suit after the site provided proof of such screening.

Hours after same-sex Army wife Ashley Broadway was named Fort Bragg's 2013 ?spouse of the year,? the on-base spouses club???that has for two months rebuffed Broadway's bid to join?? fully reversed course and invited her "to become a full member," according to emails sent to NBC News and Broadway.

The decision comes one week after the Association of Bragg Officers' Spouses (ABOS) extended Broadway???who is married to Army Lt. Col. Heather Mack???a "special guest membership," an invitation she declined and called "extremely demeaning."


"After further reviewing the (club's) constitution, by-laws and internal procedures, the ABOS Board felt that in order to immediately support all military Officer spouses who are eligible for ABOS membership a more inclusive definition of spouse was needed. Therefore, any Spouse of an active duty commissioned or warrant Officer with a valid marriage certificate from any state or district in the United States is eligible for ABOS membership," the club's board said in a statement.

"ABOS does not discriminate based on race, gender, religion, national origin, age, disability, creed, or sexual orientation. ABOS would like to publicly invite Ms. Broadway to apply for full membership to ABOS. It is and always has been our mission to support all military families."

In an email to Broadway ? shared with NBC News???the club said, "We would like to offer you to become a full member of ABOS."

"I will go ahead and submit my application," Broadway said in response to the invitation. "I need to educate some of the naysayers that are in that group and show them my family is just like their family."

In the online election held Tuesday, Broadway captured the Fort Bragg vote ?by a country mile,? said Babette Maxwell, founder of Military Spouse magazine and the Military Spouse of the Year award. Ballot totals were not revealed.

As one of the 154 base-level winners, Broadway now is eligible to be nominated for Army ?spouse of the year.?

The meeting was not granted.

?That was the most frustrating thing,? she said.

Before its decision late Friday to relent and offer Broadway full membership, the ABOS board had maintained Broadway was never rejected because ?a formal application was never filed,? and that she simply had inquired about the eligibility of a same-sex spouse and was told the club would need ?time to look at the issue.??

Online voting for the next round of the 2013 Armed Forces Insurance Military Spouse of the Year???the branch level???will take place Feb. 5. The overall winner, elected from the branch finalists, will be revealed May 9.

"I never thought in a million years I would be the one to advance the cause. If that?s what it?s going to take to get attention for all the military same-sex spouses, then so be it," Broadway said. "But I do take this (Bragg 'spouse of the year' award) very seriously. And we'll see where it goes from here."?

Immune cell suicide alarm helps destroy escaping bacteria

Cells in the immune system called macrophages normally engulf and kill intruding bacteria, holding them inside a membrane-bound bag called a vacuole, where they kill and digest them.

Some bacteria thwart this effort by ripping the bag open and then escaping into the macrophage's nutrient-rich cytosol compartment, where they divide and could eventually go on to invade other cells.

But research from the University of North Carolina School of Medicine shows that macrophages have a suicide alarm system, a signaling pathway to detect this escape into the cytosol. The pathway activates an enzyme, called caspase-11, that triggers a program in the macrophage to destroy itself.

"It's almost like a thief sneaking into the house not knowing an alarm will go off to knock down the walls and expose him to capture by the police," says study senior and corresponding author Edward Miao, PhD, assistant professor of microbiology and immunology at UNC. "In the macrophage, this cell death, called pyroptosis, expels the bacterium from the cell, exposing it to other immune defense mechanisms."

A report of the research appears online in the journal Science on Thursday January 24, 2013.

Miao, also a member of the UNC Lineberger Comprehensive Cancer Center, says the new findings show that having this detection pathway protects mice from lethal infection with the type of vacuole-escaping Burkholderia species: B. thailandensis and B. pseudomallei.

Both are close relatives. But they differ in lethality. B. pseudomallei is potentially a biological weapon. Used in a spray, it could potentially infect people via aerosol route, causing sickness and death. Moreover, it also could fall into a latent phase, "essentially turning into a 'sleeper' inside the lungs and hiding there for decades," Miao explains. In contrast, B. thailandensis, which shares many properties with its species counterpart, is not normally able to cause any disease or infection

These environmental bacteria are ubiquitous throughout S.E. Asia, and were it not for the caspase-11 pathway defense system, that part of the world could be uninhabitable, Miao points out.

This grim possibility clearly emerged in the study. Mice that lack the caspase-11 detection pathway succumb to infection not only by B. pseudomallei, but also to the normally benign B. thailandensis. "Thus caspase-11 is critical for surviving exposure to ubiquitous environmental pathogens," the authors conclude.

Miao points to research elsewhere showing that the pathway's abnormal activation in people with septic shock, overwhelming bacterial infection of the blood, is associated with death. "We discovered what the pathway is supposed to do, which may help find ways to tone it down in people with that critical condition.

As to bioterrorism, the researcher says it may be possible to use certain drugs already on the market that safely induce the caspase-11 pathway. "Since this pathway requires pre-stimulation with interferon cytokines, it is conceivable that pre-treating people with interferon drugs could ameliorate a bioterror incident. This could be quite important in the case of Burkholderia, since these bacteria are naturally resistant to numerous antibiotics.

"But first we have to find out if they would work in animal models, and consider the logistics of interferon stockpiling, which are currently cost prohibitive."

Obama Climate Change Vow Could Make EPA a Political Target

President Obama?s?second inaugural address?left no doubt about his desire to put climate change front and center in his second term. He?s likely to pursue his agenda through executive actions rather than legislation, at least initially.

That could put the Environmental Protection Agency at the center of a political battle, as Senate Republicans use the process of confirming a new head of the agency as a chance to weigh in on or even block what they see as regulatory overreach by the administration in pursuit of climate change goals.

Republicans have bristled in the last four years at regulations coming out of Obama's EPA -- Rep. Michele Bachmann of Minnesota wanted to shut the agency down altogether. Administrator Lisa Jackson was a particular target, criticized for her implementation of Clean Air Act regulations and, more recently, for conducting official business using an email alias.?

As Obama's second term begins, the EPA might become an even bigger target as Republicans brace for the likelihood that Obama will use the agency's powers to pursue his climate agenda. Environmental groups have urged the administration to use EPA's authority under the Clean Air Act to limit the carbon emissions power plants are allowed to produce and to implement stricter standards on leaks of methane, a greenhouse gas, both of which can be achieved without any further congressional approval.

Although EPA is the prime symbol of the Obama administration?s climate change agenda, other agencies will also play a role?agencies that are expected to face confirmation fights in the coming months. The Energy Department, for example, can adopt new efficiency standards for home appliances. The State Department will rule on a permit for part of the Keystone XL pipeline that would cross into Canada. Each of those agencies is?likely to face or already in the process of confirming a new director, and questions over their views on climate could be part of the confirmation hearings. At EPA, Jackson announced she was stepping down last month, and the administration has yet to nominate a replacement.

Congress isn?t expected to take up climate legislation any time soon. It will be preoccupied in coming months by a series of looming budget fights, gun control legislation, and immigration reform. A sweeping cap and trade bill that Obama had pushed in his first term never made it through Congress. This time, Senate Democrats?have indicated?they are ready to let the EPA run the show.

So get ready for a show.?

Melinda Pierce, deputy director for national campaigns at the Sierra Club, said the hearing to replace Jackson at EPA might be "doubly contentious" if the agency is perceived as a key stakeholder in moving Obama's climate agenda forward.?

The focus on climate in Monday's inaugural speech surprised?and pleased?environmental groups. But it could also set up a tougher hearing, said Andrew Wheeler, a former Republican staff director and chief counsel for the Senate Environment and Public Works Committee.

?During the campaign, President Obama tried to appeal to the coal states. He never once mentioned climate change. He tried at one point to even go to the right of Mitt Romney on coal issues. He didn?t mention climate change until the night of the election after he won. In his acceptance speech, he mentioned climate change and then he mentioned it again this week.... I think that?s going to generate a lot of questions and concerns from members, not just Republicans, but also moderate Democrats,? Wheeler said.?Especially vulnerable are the six Democratic senators from red states who will face tough reelection battles in 2014.

What?s more, Wheeler said, now that Obama is in his second term, there won?t be the typical deference given to a new president?s Cabinet selections.

Confirmation hearings are a good place for the minority to be heard on administration goals. ?You?ll get good press out of it and, quite frankly, you might not have the administrator come back up to the Hill until the following year,? said David Banks, a former deputy staff director for Republicans on the Senate Environment and Public Works Committee.

But even if the EPA confirmation fight is nasty, it probably would not deter Obama from implementing his second-term climate agenda. Under the Clean Air Act, the EPA has a legal duty to issue regulations limiting power-plant carbon emissions, said Conrad Schneider, advocacy director at the Clean Air Task Force, which recently sent an open letter to Obama outlining its recommendations for addressing climate change over the next four years. CATF also believes that the EPA failed to do its duty with respect to regulating emissions of methane from the oil and gas industry during Obama?s first term. ?Since those are statutory duties, it really doesn?t matter who is at the head of the agency. Those duties exist if the nominee is confirmed quickly and those duties exist if the nominee?s confirmation process drags out,? he said.

So consider the confirmation hearings a preview of the bigger fight on climate change in Congress over the next four years. Republicans may try to block the new regulations. Obama may try to get another climate bill through Congress. Legislation, environmental groups say, is the best way for Obama to secure the climate-change legacy he wants in the long run.
